First for Please Teacher!, when I noticed the Japanese voice of Mizuho who addresses Kei would always sound like she is saying "Cake"? But why is pronounced that way for the Japanese version?

Also, most of the animes I watch with female characters wearing eyeglasses, I noticed that when they are overexcited, the lens on their glasses brightens. I felt like I have seen an illustration of that just like the one I saw for a character in Ranma 1/2 named Mousse. But at that time, I was only playing the video game version. Take Please Twins! and Fullmetal Alchemist for example, Tsubaki Oribe and Sciezka, both wear eyeglasses and when excited, their glasses brightens. There must be some explanation of this matter. It would help with an answer.

She's probably saying "Kei-kun". I'm not familiar enough with it's specific usage to explain it properly, so I'll quote Wikipedia:

Wikipedia wrote:
Kun (君) is an informal and intimate honorific primarily used towards males (It is still used towards females, but rarely.) It is used by persons of senior status in addressing those of junior status, by males of roughly the same age and status when addressing each other, and by anyone in addressing male children. In business settings, women, particularly young women, may also be addressed as kun by older males of senior status. It is sometimes used towards male pets as well.

Schoolteachers typically address male students using kun, while female students are addressed as san or chan. The use of kun to address male children is similar to the use of san when addressing adults. In other words, not using kun would be considered rude in most situations, but, like san for members of one's own family, kun is traditionally not used when addressing one's own son (unless kun is part of a nickname: "Akira-kun"—Akkun) or when referring to one's own child in conversations with others.

At times that's to make the character seem funny, and most people (in Japan) will know what kind of mood that person is in. It's not just female characters though, it happens to male characters as well. To show how smart or how dumb they might be, or whether they've just come up with a great idea that could be the plot of the episode, or their downfall. Just how it comes.
