Uhhhh, I don't know anything about live action Japanese television, but I'm assuming the large amount of nudity-based humor won't be used here?

I can only imagine this working as very tongue in cheek. A big part of their character is meant to be that they are identical, six same faces.
The series itself would probably make a joke of the idea of live action where they suddenly have different facial features from each other.

Fairly sure every single manga to live-action adaptation are never identical.
From Kimi ni Todoke, Nodame Cantabile, Lupin III, Detective Conan, Kindaichi Case Files, and more. And average watcher in Japan and other Asian countries who are prominent market for Japanese drama doesn't care at all.

Oh, they do care. Half of the Twitter reactions is "lol, but they're supposed to be identical" (well, that, and also "WHY" and "PLEASE NO") and it makes sense, considering that the entire franchise is based on the characters being, you know, sextuplets.
But this movie is not targeting fans of Osomatsu-san, it's targeting those, fans of the idol group, and other people who just want to see pretty boys on screen being cute and funny, and well, those people are likely not familiar with Osomatsu-san, and thus are not going to care.

I think you missed the point. The main characters of this anime are physically identical sextuplets. That's the central joke.
